چکیده

Cultivation-independent genomic approaches have greatly advanced our understanding of the ecology and diversity of microbial communities involved in biodegradation processes. However, much still needs to be resolved in terms of the structure, composition and dynamics of the microbial community in impacted ecosystems. Here we report on the RNA activity of the microbial community during the bioremediation process using RNA Temperature Gradient Gel Electrophoresis (RNA-TGGE). Dendrograms constructed from similarity matching data produced from the TGGE profiles separated a community exhibiting high remediation potential. Overall, increased Shannon Weaver Diversity indices (1–2.4) were observed in the high potential remediation treatment samples. The functionality of the microbial community was compared, with the microbial community showing the greatest organisation also showing the highest levels of hydrocarbon degradation. Subsequent sequencing of excised bands from the microbial community identified the presence of Gammaproteobacteria together with a number of uncultured bacteria. The data shows that RNA TGGE represents a simple, reproducible and effective OPEN ACCESS J. Mar. Sci. Eng. 2015, 3 969 tool for use in the assessment of a commercial bioremediation event, in terms of monitoring either the natural or augmented hydrocarbon-degrading microbial community.

Environmental Monitoring through Next- Generation Sequencing Metabarcoding: As- Sessing the Impact of Fish Farming on Benthic Macro-and Meiofauna Communities

The measurement of species diversity represents a powerful tool for assessing the impacts of human activities on marine ecosystems. Traditionally, the impact of fish farming on the coastal environment is evaluated by monitoring the dynamics of macrobenthic infaunal populations. However, taxonomic sorting and morphology-based identification of the macrobenthos demand highly trained specialists a...

Degradation of Alkanes in Contaminated Sites

Petroleum hydrocarbons are important energy resources used by industry and in our daily life, whose production contributes highly to environmental pollution. To control such risk, bioremediation constitutes an environmentally friendly alternative technology that has been established and applied. It constitutes the primary mechanism for the elimination of hydrocarbons from contaminated sites by ...

Cumulative Human Impacts on Mediterranean and Black Sea Marine Ecosystems: Assessing Current Pressures and Opportunities

Management of marine ecosystems requires spatial information on current impacts. In several marine regions, including the Mediterranean and Black Sea, legal mandates and agreements to implement ecosystem-based management and spatial plans provide new opportunities to balance uses and protection of marine ecosystems. Analyses of the intensity and distribution of cumulative impacts of human activ...

Characterization of naphthalene-degrading bacteria isolated from the Persian Gulf and the Caspian Sea as potential agents for naphthalene removal from polluted environments

Over fifty bacterial strains were isolated from seawater samples in the presence of naphthalene as a sole source of carbon and energy. Among them, three isolates with higher growth rate and naphthalene degradation ability were selected for further studies. Biochemical and molecular analysis revealed that two Persian Gulf isolates, strain PG-10 and strain PG-48 belonged to the group of hydrocarb...
